Kloniranje i ekspresija
urediSekvenciranjem klonova dobijenih iz biblioteke cDNK ljudskog mozga frakcionirane veličine, Nagase et al. (1998) klonirali su ZC3H13, koji su označili kao KIAA0853. Izvedeni protein od 967 aminokiselina ima slabu homologiju s ljudskim trihohialinom (TCHH). RT-PCR ELISA testom otkrili je najjaču ekspresiju KIAA0853 u jetri, jajnicima i mozgu, zatim u bubrezima. Umjerena ekspresija otkrivena je u srcu, plućima, skeletnim mišićima, gušterači i sjemenicima.[8]
Funkcija gena
urediKoristeći masenu spektrometrijsku analizu za identifikaciju proteina koji su imunoprecipitirani sa WTAP iz endotelnih ćelija ljudske pupčane vene (HUVEC), Horiuchi et al. (2013) identificirali su proteinski kompleks, koji je uključivao KIAA0853, HAKAI (CBLL1), virilizer (KIAA1429), RBM15, BCLAF1 i THRAP3. WTAP kompleks lokaliziran na jedarnim pjegama HeLa ćelija i HUVEC-ovima, a eksperimenti umrežavanja pokazali su da je prolazno povezan s faktorima prerade. Nokauniranje bilo koje komponente WTAP kompleksa, uključujući KIAA0853, smanjilo je alternativnu preradu WTAP-a, što je rezultiralo povećanom proizvodnjom proteina pune dužine WTAP-a i smanjenom proliferacijom ćelija, sa zastojem ćelijskog ciklusa u fazi G2. Razgradnja BCLAF1 i THRAP3 smanjila je lokalizaciju jedarnih pjega drugih komponenti WTAP kompleksa.[9]
