Kloniranje i ekspresija
urediSekvenciranjem nasumično odabranih cDNK, koje odgovaraju relativno dugim transkriptima iz ljudskog mozga, Nagase et al. (1997) identificirali su djelimičnu SFRS14 cDNK, koju su nazvali KIAA0365.
Pretragom baze podataka EST i analizom genomskih sekvenci, Sampson i Hewitt (2003) identificirali su SFRS14 u punoj dužini, kao i povezanog člana porodice SURP, SF4 (SUGP1). SFRS14 kodira izvedeni protein od 1,083 aminokiseline, s procijenjenom molekulskom masom od 120,2 kD. I SF4 i SFRS14 sadrže 2 SURP motiv i jedna domen G-plohe na svojim C-krajevima. SFRS14 također sadrži N-kraj bogat argininima /serinima, što sugerira da je novi član porodice pre-iRNK faktora prerade povezanih sa SR.
Sampson i Hewitt (2003) također su klonirali mišje ortologe SF4 i SFRS14. Mišji protein Sfrs14 ima identičnu organizaciju domena i 73% identiteta sekvence sa svojim ljudskim homologom. Northern blot analiza otkrila je ekspresiju otprilike 3,5 kb SFRS14 transkripta samo u sjemenicima i fetusnom mozgu i bubrezima. RT-PCR identificirao je ekspresiju SFRS14 u skeletnim mišićima i mozgu odraslih.
Funkcija gena
urediNa temelju prisutnosti domena SURP, G-plohe i RS, Sampson i Hewitt (2003) sugeriraju da SFRS14 funkcionira u mehanizmima prerade preiRNK.
Struktura gena
urediSampson i Hewitt (2003) utvrdili su da gen SFRS14 sadrži 12 egzona i da se proteže na 41,2 kb. SFRS14 podvrgava se [[alternativna prerada RNK|alternativnoj 3-osnovnoj preradi. Najvećem SFRS14 transkriptu nedostaje egzon 11, koji čini prva 103 bazna para UTR-a s tri prajmera.
Mapiranje
urediHibridnom analizom zračenja Nagase et al. (1997) mapirali su gen SFRS14 na hromosomu 19. Analizom sekvence, Sampson i Hewitt (2003) mapirali su gene SFRS14 i SF4 unutar 300 kb, jedan od drugog, na hromosomu 19, pozicija na kratkom kraku: p13.11.[7][8]
