Kloniranje i ekspresija
urediPregledavanjem ljudske genomske biblioteke mišje cDNK Mybbp1a, Keough et al. (1999) izolirali su dio ljudskog gena MYBBP1A, koji su također nazvali P160. Koristeći genomsku sekvencu, klonirali su ljudsku cDNK MYBBP1A pune dužine. Predviđeni 1.328-aminokiselinski MYBBP1A sadrži centralno kiselo područje, ponavljanja C-terminalnih baznih aminokiselina i sekvencu potencijalnih motiva leucinom nabijenih domena u proteinu. Ljudski MYBBP1A dijeli približno 80% sličnosti aminokiselinskih sekvenci s mišjim i miševima Mybbp1a. Northern blot analizom otkriven je približno 4,5 kb MYBBP1A transkript u svim ispitivanim ljudskim linijama, a to su epiteli bubrega, eritroleukemijski karcinomi i grlića maternice, te ćelijske linije fibroblasta. Otkrili su da se ljudski EST koji odgovaraju MYBBP1A eksprimiraju u širokom spektru tkiva, što sugerira da se MYBBP1A eksprimira sveprisutno.[8]
Funkcija gena
urediProtoonkogen MYB pretežno se eksprimira u nezrelim hematopoetskim ćelijama, gdje ima važnu ulogu u proliferaciji i diferencijaciji hemopoetskih ćelija. Onkogeno aktivirani oblici MYB su obično imaju N– i / ili C-terminalne delecije normalnog MYB proteina. Uklanjanje C-kraja MYB remeti ili briše regiju nazvanu negativni regulatorni domen (NRD), što rezultira povećanjem vezivanja, transaktivacije i transformacije DNK od pomoću MYB-a. Jedna od karakteristika NRD-a je motiv poput leucinskog zatvarača. Tačkaste mutacije koje remete ovaj motiv također pojačavaju potencijal transaktivacije i transformacije MYB-a. Protein-1a koji veže mišji Myb (Mybbp1a), izvorno nazvan P160, identificiran je njegovom sposobnošću specifične interakcije s Myb, putem ovog motiva sličnog leucinskom zatvaraču. Zato je predloženo da Mybbp1a može modulirati MYB aktivnost nakon vezanja za MYB NRD. Mybbp1a je sveprisutno eksprimiran.[9][10]
